Welcome to the Winogradsky column lab page! Cadiz, 9B: Winogradsky column, first week.



Hi, everyone!  We are three students from UCA
The sources of the mud that we used are the river and the beach,  salt water was added to the mixture. We've mixed the mud with paper, calcium carbonate , ferrous sulfate and calcium sulfate.
The paper was added to the mixture due to the fact that it provides a source of carbon(cellulose), the calcium carbonate was used because it produces carbon dioxide, another source of carbon. Ferrous sulfate and calcium sulfate were used as a source of sulphur.
The Winogradsky column was made on Tuesday 3rd march 2015.
The mud was mixed with the carbon and sulphur sources in a bucket, then, it was added to the bottle and packed in the bottom. More mud was added to fill two thirds of the bottle capacity. Then the salt water was  added to the bottle.
The column has been placed next to a window.
The added material triggered microbial activity that depletes the oxygen in the mud, creating an oxygen gradient in the column. Under these anaerobic conditions, anaerobic bacteria like Clostridium predominate. These bacteria use the cellulose from the paper to obtain glucose subunits. After obtaining glucose, the bacteria partially break it down by fermentation, producing by-products like ethanol and organic acids.
These by-products feed into other bacteria. Desulfovibrio use the organic acids as a carbon source, they also use the sulfate that was added to the column(desulfovibrio bacteria use sulfate as a final electron receptor in respiration, producing hydrogen sulfide.).
The hydrogen sulfide is used by the green sulphur bacteria and the purple sulphur bacteria, which will also use carbonic dioxide obtained from calcium carbonate.
The presence of iron bacteria may explain the orange coloured water. The black dots are probably desulfovibrio bacteria,  the foul odour of the column is caused by them. Finally, the brown colour can be explained by the presence of anaerobic bacteria.

Cadiz, 6B: Winogradsky column: First week (Group 6B)

Hello winobloggers,
We apologize for the delay. This is our first post about the Winogradsky column. We are other three students of biotechnology in the University of cádiz.

We made our Winogradsky column on tuesday, 3th March in our Microbiology laboratory. Our column contains a mixture of muddy sediment from San Pedro river and muddy sediment from a hypersaline pond. Moreover, we added as enrichments : CaCO3, FeSO4 and NaCl(salt).

First, we mixed  the sediment from the river and from the pond in a bucket.We put more sediment from the river and we mixed with our hands. Then, we added the enrichment (one teaspoon of each compounds) and we kneaded again. After that, we introduced the mix in a bottle (around two thirds) and finally, we poured 2 cm of water from the same river.
Our column is situated in a desk next to a window, where receives  the sunlight only on one side.
The column has changed much in this week. The first day it has a grey colouration, the water was turbid and it had two worms. However the tuesday, 10th march, the water was yellowish, the column was dark grey with a reddish colouration on the side where receives the sunlight and black spots have appeared.

We think that  in our Winogradsky column are living or will appear: cyanobacterias in the water (the aerobic zone);purple and green sulfer bacterias in the middle; sulfate-reducing bacteria in the bottom because we added FeSO4 and these bacterias producing a nauseating smell and the black spots that we can see; and ,maybe, halophile bacterias because we added salt. In summary, we think that in the future our Winogradsky column will probably be similar to the theoretical image.

Cadiz, 5B: winogradsky column 5B group

Hello, this is our first post in this blog. We are students from Cádiz University currently in the 1st year of Biotechnology.
For our Winogradsky column we have used: soil from the ground and the lagoon and sandy beach sediment taken from Rio San Pedro. It has been added material enrichment such as calcium sulfate, pieces of filter paper, motor oil and calcium nitrate. This was all mixed inside a bucket and placed in a bottle using a funnel, filling 3/4 of the Aquarius bottle with it then we added water from the tab to complete 4/5 of the Aquarius bottle. The column was created on tuesday , 3rd March in one the science laboratories of the faculty.
The bottle is placed on a balcony in Cádiz where the day temperatures are high and the nights temperatures are low, it also  only gets sun on one side, while the other side is facing the wall.
Our hypothesis is that in the top of the bottle grow aerobics microorganisms, and at the end grow microorganisms that do not need a lot of oxygen to live (anaerobics). The side of the bottle that receives sun, has green spots due to the appearence of photosynthetic microorganisms and in the side facing the wall there are black and big spots. The presence of paper make microorganisms that need carbon grow; with calcium sulfate, sulfate reducing microorganisms grow and with the calcium nitrate denitrifying microorganisms grow.

Cadiz, 7B: Experiment of Winogradsky column. First week(3th -10th march)

In this blog, I talk about the observations of the Winograndsky´s column which my group are doing. We are two students of biotechnology in the university of Cadiz.

The Winograndsky´s column contains sand of the beach and mud of the river San Pedro that take up around the half of the bottle and a quarter of water from the same river. The compounds, that we added like enrichment were CaSO4, FeSO4, CaCO3 and starch. The bottle is located in a window. In this place, the sun warm up only a part of the bottle.

On Tuesday, 3th March ,The column were created in a laboratory of the university. At the beginning, the bottle have a grey colouration and the water was turbid. In the third day, we could see that there were black spots and white lines in the bottle and the water was yellow. After a few days, we observed that an orange line is formed in the walls of bottle that is in contact with the water. The mixture smells bad.

If we used these observations, we could deduce that in the column  there are :

-Cyanobacteria in the surfaces of the bottle because they are aerobics.
-Bacteria that reduces the ions sulphates of CaSO4 and FeSO4 to sulphides. They are the black spots that we can see. Other way of detected them is by the smell. The compound H2S(product of reaction) segregates a strong smell to decomposed eggs.
-Bacteria fermenting glucose because we added starch.
- Bacteria of iron that produces reaction of oxidation in this element . This type of bacteria changes the colour of water to yellow or orange and they can improve the proliferation of the reducing-sulphates bacteria.

Cadiz 1A: WINOGRANDSKY COLUMN, FIRST WEEK RESULTS (GROUP 1-A)

    Hello Winoblog, we are three students from Cádiz University and this is the first post we publish about our Winograndsky column.
    We have used sandy beach sediment from Río San Pedro beach. The column was created on Wednesday, 4th March, in Science Faculty laboratory.
    It has been added material enrichment such as calcium sulfate, calcium carbonate, filter paper cut into pieces (cellulose), and sodium chloride (salt).
    Firstly, it has been added sandy beach sediment on a bucket. Then, it has been added the material enrichment mentioned before and meanwhile we were mixing at the same time. Before the mixture was introduced in the Aquarius bottle (Winograndsky column), it had been added a little bit of watter from the same beach as the sediment, so that the mixture got more liquid than before. Finally, once the mixture was introduced in the bottle, it was added two centimetres more of watter.
The sediments take up 0.8 dm3 of the bottle.
    The column is located next to a window. The sun warms up one side of the column and the other side doesn't receive the sun light because it is next to a wall.
    If we analyze the progress reached in the first week, our bottle contains black points spread out uniformly. We notice black lines in the part which the sun warms up. There are bigger black points which are probably colonies formed by bacterias. There are some dark spots (they're grey) too. That spots are more numerous and bigger in the dark side of the bottle. The last phenomenon we can observe is that, in the surface, there are tiny shells sticked to the bottle.
    The scientific hypothesis is that it will probably appear purple and green bacterias (due to we have added sulphates), cyanobacterias and seaweeds in the surface of the bottle (due to they're aerobic bacterias), and grey and dark grey bacterias (sulfur-reducing bacterias and glucose fermenting bacteria, respectively).

UCA team joins the Winoblogger world!


Dear Winobloggers,

Thank you for the invitation to participate in your blog.

We have spent the last couple of days preparing our Winogradksy columns. We have 29 columns ready.

We have used sandy beach sediment, muddy sediment from a tidal creek and muddy sediment from a hypersaline pond. Some of us have also prepared nice cocktails of all three sediments.

We were not frugal neither with the stuff we added: CaCO3, CaSO4, FeSO4, filter paper (cellulose), starch, NaCl, and combinations of the above. Some even added some motor oil!

I attach a map to show you where we collected the sediment from.

Last but not least...

    Η κατασκευή της στήλης πραγματοποιήθηκε στις 17/10/14 στον ποταμό Λούρο. Σε ένα άδειο πλαστικό μπουκάλι, το οποίο κλείνει με πώμα, προστέθηκαν ως υλικά εμπλουτισμού περίπου 20 μικρά κομμάτια από εφημερίδα ως πηγή άνθρακα και ένα ολόκληρο αβγό, βρασμένο για 5 λεπτά, μαζί με το τσόφλι, ως πηγή θείου και ασβεστίου. Αρχικά προστέθηκε λίγη λάσπη από την όχθη του ποταμού για ανάμιξη με τα υλικά εμπλουτισμού και στην συνέχεια προστέθηκε και άλλη λάσπη, έως ότου να καλυφθεί το μπουκάλι περί τα 2/3. Στην συνέχεια προστέθηκε νερό για την κάλυψη του μισού του ήδη υπάρχοντος κενού της στήλης. Τέλος στο μπουκάλι προστέθηκε το βιδωτό πώμα ώστε να μην υπάρχει επικοινωνία του εσωτερικού με το εξωτερικό της στήλης. Η στήλη τοποθετήθηκε στο μπαλκόνι, αφού οι καιρικές συνθήκες το επέτρεπαν(καλή θερμοκρασία, αρκετός ήλιος).

1η μέρα: άρχισε να σταθεροποιείται, το νερό ήταν διαυγές καθώς χωρίστηκαν πλήρως νερό και λάσπη.
   

     
6η μέρα: η στήλη δεν παρουσιάζει φαινοτυπικές αλλαγές αλλά περνάει από το μπαλκόνι μέσα στο σπίτι, γιατί σιγά σιγά οδεύοντας προς το Νοέμβρη πιάνουν τα πρώτα κρύα(χαμηλή θερμοκρασία, λιγότερος ήλιος). Μέσα στο σπίτι τοποθετείται σε σημείο που το βλέπει το αμελητέο πλέον φως του ηλίου αλλά και το φως της λάμπας. Όλες αυτές τις μέρες παρατηρούνται πολλές φουσκάλες στο πάνω μέρος της στήλης, είναι το εγκλωβισμένο οξυγόνο που βγαίνει στην επιφάνεια. 








20η 
μέρα: αρχίζει δειλά δειλά και εμφανίζεται ένα κοκκινωπό χρώμα στο επάνω μέρος της λάσπης, πιθανώς πρόκειται για την ύπαρξη πορφυρών θειοβακτηρίων, ενώ ταυτόχρονα παρατηρείται η εμφάνιση μαύρου χρώματος στα κατώτερα στρώματα της λάσπης, που είναι τα θειοαναγωγικά βακτήρια. 

109η μέρα:  

Προηγήθηκαν

  • 20 μέρες Χριστουγέννων, όπου η θερμοκρασία στο σπίτι ήταν άκρως χαμηλή (περίπου 9oC) και δεν υπήρχε καθόλου φως
  • ένα αναπάντεχο αναποδογύρισμα της στήλης, που "εξαφάνισε" το κόκκινο στρώμα των πορφυρών θειοβακτηρίων (λογικά διασκορπίστηκαν στα τοιχώματα του μπουκαλιού). 
     
                                                                                                                                    Παρατηρήσεις:
  • δεν υπήρχε καθόλου ανάπτυξη κυανοβακτηρίων και πράσινων θειοβακτηρίων, κάτι που μπορεί να οφείλεται στην κακή αναδευση των υλικών εμπλουτισμού, αφού ένα βρασμένο αβγό δεν αναμιγνύεται το ίδιο εύκολα με ένα ωμό!
  • ιδιαίτερη εντύπωση μου προκάλεσε το γεγονός οτι την 109η ημέρα προσπάθησα να το φωτογραφίσω στο ίδιο ράφι την 20η ημέρα αλλά παρατήσηρα ότι η στήλη πλέον δεν χωρούσε. Από την στιγμή που το ράφι είχε ακριβώς τις ίδιες διαστάσεις ή το πλαστικό μπουκάλι ήταν αυτό που διογκώθηκε λόγω της υψηλής παραγωγή Η2S, ή εγώ αρχίζω να εμφανίζω σημάδια παράνοιας...

Καλησπέρα.

Για την κατασκευή της στήλης Winogradsky η δειγματοληψία έγινε στον ποταμό Λούρο στις 17/10/14. Η στήλη αποτελείται από λάσπη και νερό από τον ποταμό. Σε ένα πλαστικό μπουκάλι νερού αρχικά προστέθηκαν τα υλικά εμπλουτισμού. Ως πηγή άνθρακα χρησιμοποιήθηκε εφημερίδα και ως πηγή θείου ωμό αυγό μαζί με το τσόφλι το οποίο περιέχει ανθρακικό ασβέστιο. Στη συνέχεια προστέθηκε λάσπη. Το υπόλοιπο της στήλης καλύφθηκε με νερό από το ποτάμι αφήνοντας ένα χώρο με αέρα πάνω από την επιφάνεια του. Μέτα την διαδικασία παρασκευής της στήλης αυτή τοποθετήθηκε σε σημείο όπου δεν υπάρχει υψηλή έκθεση στον ήλιο ή άλλη πηγή φωτός και αυτό φτάνει μόνο εξ αντανακλάσεως. 

Μετά από 3 βδομάδες η εικόνα της στήλης είναι η παρακάτω στην οποία διακρίνεται μια ελαφρώς καφέ περιοχή στο πάνω μέρος της λάσπης: 


 
Σε λιγότερο από τρεις μήνες παρουσιάστηκε το παρακάτω αποτέλεσμα:



Μέσα σ'αυτό το χρονικό διάστημα υπήρξαν περίπου 20 μέρες όπου δεν υπήρχε καθόλου φως. Παρατηρείται ένα κόκκινο χρώμα στο νερό της στήλης που οφείλεται στα πορφυρά θειοβακτήρια. Επίσης υπάρχει και ένα μαύρο χρώμα στο κατώτερο τμήμα που υποδηλώνει την ανάπτυξη των θειοαναγωγικών βακτηρίων. Τα κυανοβακτήρια και πράσινα θειοβακτήρια δεν έκαναν την εμφάνιση τους μάλλον λόγω χαμηλής έντασης φωτός που έχει ως συνέπεια τον αργό ρυθμό ή και καθόλου ανάπτυξης τους.
