PREPARATION OF WINOGRADSKY COLUMN

16/03/2017

The Winograsky column constitutes a model of aquatic ecosystem where we can see the bacterial photosynthesis and the cycles of the sulfur produced by the metabolisms of the microorganisms that proliferate in it.

In our case, we made two columns from 60 grams of mud, 220 grams of sand and sea water. To prepare columns, we began by preparing an enrichment mixture with nutrients favorable for the development of microorganisms. This medium consisted of 1 gram of gypsum (calcium sulphate), since it is a source of calcium and sulfur, and 0.54 grams of dipotassium phosphate as it provides potassium and phosphorus.

Once this mixture was prepared, we separated in equal parts to created two columns which would be subject to different environmental conditions: one will be under direct sunlight conditions and the other one in darkness. After a few weeks we will return to visualize and comparte columns in order to observe if there are any differences between them.

In the next image we could see two columns, the one on the left was placed in the dark, and the one on the right was put in sunlight.

  1. So what is your hypothesis? Why did you use the specific compounds?
